چکیده

6-Hydroxydopamine (6-OHDA) is one of the most widely used rat models for Parkinson's disease. There is ample evidence in the literature that 6-OHDA elicits its toxic manifestations through oxidant stress. In the present study, we evaluated the anti-parkinsonian effects of Withania somnifera extract, which has been reported to have potent anti-oxidant, anti-peroxidative and free radical quenching properties in various diseased conditions. Rats were pretreated with 100, 200 and 300 mg/kg b.w. of the W. somnifera extract orally for 3 weeks. On day 21, 2 microL of 6-OHDA (10 microg in 0.1% in ascorbic acid-saline) was infused into the right striatum while sham operated group received 2 microL of the vehicle. Three weeks after 6-OHDA injections, rats were tested for neurobehavioral activity and were killed 5 weeks after lesioning for the estimation of lipidperoxidation, reduced glutathione content, activities of glutathione-S-transferase, glutathione reductase, glutathione peroxidase, superoxide dismutase and catalase, catecholamine content, dopaminergic D2 receptor binding and tyrosine hydroxylase expression. W. somnifera extract was found to reverse all the parameters significantly in a dose-dependent manner. Thus, the study demonstrates that the extract of W. somnifera may be helpful in protecting the neuronal injury in Parkinson's disease.

Evaluation of Betaine Neuroprotective Effects on 6-Hydroxydopamine-Induced hemi-Parkinsonism in Male Wistar Rats

BACKGROUND: Parkinson's disease (PD) is one of the prevalent debilitating neurodegenerative disor- ders. Accordingly, researchers are working on methods to modify PD progression. Previously, the neuro- protective effects of betaine, as a methyl donor agent in homocysteine metabolism, have been demonstrated in animal models of chronic cerebral hypoperfusion and memory deficits. OBJECTIVES: It wa...

Research Paper: The Anti-Parkinsonism Effects of KATP Channel Blockade in the 6-Hydroxydopamine-Induced Animal Model: The Role of Oxidative Stress

Introduction: Studies suggest that ATP-sensitive potassium (KATP) channels are a potential pharmacotherapeutic target for neuroprotection in neurodegenerative diseases. The current study aimed at evaluating the effect of pretreatment with glibenclamide (Glib) and B vitamins supplement on the severity of behavioral symptoms in 6-hydroxydopamine (OHDA)-induced Parkinsonism. Also malondialdehyde (...

B Vitamins Supplement Potentiates Antiparkinsonian Effect of Flunarizine: the Behavioral and Biochemical Evidences From 6-Hydroxydopamine Animal Model

Introduction: Prominent data indicate that flunarizine (flu), a calcium channel blocker, has neuroprotective effect. However, several authors have reported that the chronic use of flu can produce drug-induced Parkinsonism. Previously, we showed that B vitamins supplement (B com) has antiparkinsonian effect. In the present study, we evaluated the effect of pretreatment with flu ...

Phytoremedial effect of Withania somnifera against arsenic-induced testicular toxicity in Charles Foster rats

Objective: The main objective of the current study was to observe the ameliorative effect of Withania somnifera on arsenic-induced testicular toxicity by exploring the crucial parameters such as sperm counts, sperm motility, hormonal assay and lipid peroxidation including histopathology. Materials and Methods: In the present study, arsenic in the form of sodium arsenite was administered orally ...

Protective Effects of Withania somnifera Root on Inflammatory Markers and Insulin Resistance in Fructose-Fed Rats

Background: We investigated the effects of Withania somnifera root (WS) on insulin resistance, tumor necrosis factor α (TNF-α), and interleukin-6 (IL-6) in fructose-fed rats. Methods: Forty-eight Wistar-Albino male rats were randomly divided into four groups (n=12); Group I as control, Group II as sham-treated with WS by 62.5mg/g per diet, Group III fructose-fed rats received 10%...
